HLA typing using next generation sequencing: An overview

The introduction of PCR based amplification and sequenc- 13 ing of HLA class I and class II genes revealed a level of polymor- 14 phism far beyond the resolution of serologic or cellular based 15 HLA typing. Now, decades later, the HLA community is still strug- 16 gling to deal with this extensive and ever-expanding allelic diver- 17 sity (12,542 HLA alleles as of January, 2015). The patchwork 18 pattern of polymorphism identified by these initial sequencing 19 studies was primarily localized to the exons that encoded the pep- 20 tide binding domains of class I and class II molecules. more
