Scientists report positive data from a first-in-man study evaluating the bone marrow-protective effects of genetically modified autologous hematopoietic stem cell transplants in glioblastoma patients treated using chemotherapy and benzylguanine. The results, published in Science Translational Medicine, showed that of the three patients given the transplants prior to combination therapy, two survived much longer than average, and the third remains alive with no disease progression almost three years later. Pancreatic islet transplantation is a promising method for curing diabetes mellitus. We proposed in this study a molecularly engineered islet cell clusters that could overcome problems posed by islet transplantation circumstances and host's immune reactions. A gene containing highly releasable Exendin-4, an insulinotropic protein, was delivered into single islet cells to enhance glucose sensitivity; thereafter, the cells were reaggregated into small size ICCs.
