The February special issue of SLAS Discovery, "Advances in Cellular Target Engagement and Target Deconvolution," features 13 articles focused on multiple aspects of and technologies related to cellular target engagement. Special issue editors Martin Main, Ph.D., (Medicines Discovery Catapult, UK) and Andrew Zhang, Ph.D., (AstraZeneca, USA) selected articles that span a variety of relevant topics, from the direct measurement of compound-target interaction using BRET technologies to the measurement of compound-induced changes in protein stability and localization. Open access is available until February 20.

European Sample Management Symposium Keynote Speaker Announced
SLAS
Gisbert Schneider, Ph.D., professor and Chair for Computer-Assisted Drug Design at ETH Zurich, is a pioneer in the integration of machine-learning methods into practical medicinal chemistry. He coined the phrases "scaffold-hopping" and "frequent hitter" and his research has led him to hold many top positions in both academia and pharma. He is also confirmed as this year's European Sample Management Symposium keynote speaker (16-17 March, Berlin, Germany) where he'll present on molecules on demand and the opportunities and limitations of designing them using machine intelligence. This is a can't-miss presentation that we're very excited to host!
As a reminder, the deadline to submit a poster abstract related to supply chain management, sample collections, data management or robotics AND the opportunity to save with the Early Bird Registration rate is Monday, 10 February.

Multivalent Cation-Induced Actuation of DNA-Mediated Colloidal Superlattices
Journal of the American Chemical Society
Nanoparticles functionalized with DNA can assemble into ordered superlattices with defined crystal habits through programmable DNA "bonds." Here, we examine the interactions of multivalent cations with these DNA bonds as a chemical approach for actuating colloidal superlattices. Multivalent cations alter DNA structure on the molecular scale, enabling the DNA "bond length" to be reversibly altered between 17 and 3 nm, ultimately leading to changes in the overall dimensions of the micrometer-sized superlattice.

Predicting Breast Cancer by Paper Spray Ion Mobility Spectrometry Mass Spectrometry and Machine Learning
Analytical Chemistry
Paper spray ionization has been used as a fast sampling/ionization method for the direct mass spectrometric analysis of biological samples at ambient conditions. Here, we demonstrated that by utilizing paper spray ionization-mass spectrometry coupled with field asymmetric waveform ion mobility spectrometry, predictive metabolic and lipidomic profiles of routine breast core needle biopsies could be obtained effectively.

Chemists Teach Neural Networks to Predict Properties of Compounds

A new joint Russian-French-Japanese team has developed a computational model able to predict the properties of new molecules based on the analysis of fundamental chemical laws. The study, titled "Using AI methods for the planning of chemical synthesis," is published in the Journal of Chemical Information and Modeling.
Associate Professor Timur Madzhidov says, "We offered a way to insert the preexisting chemical equations into some frameworks of machine learning."

Walking With Atoms: Chemical Bond Making and Breaking Recorded in Action

Ever since it was proposed that atoms are building blocks of the world, scientists have been trying to understand how and why they bond to each other. Be it a molecule (which is a group of atoms joined together in a particular fashion), or a block of material or a whole living organism, ultimately, everything is controlled by the way atoms bond, and the way bonds break.
The challenge is that lengths of chemical bonds are between 0.1-0.3 nm, about half a million times smaller than the width of a human hair, making direct imaging of bonding between a pair of atoms difficult.

Protein editorial assistants are clearing the way for cut-and-paste DNA editors, like CRISPR, to access previously inaccessible genes of interest. Opening up these areas of the genetic code is critical to improving CRISPR efficiency and moving toward futuristic, genetic-based assaults on disease.
The DNA-binding editorial assistants were devised by a US-based team of bioengineers, who describe their design in APL Bioengineering, from AIP Publishing.

Immune checkpoint inhibitor drugs have added a dimension to physicians' arsenal in their fight against cancer. However, not all patients will respond to the currently approved therapies, wasting precious time that could be spent using other potentially lifesaving drugs. Successes are limited by the lack of biomarkers that identify patients most likely to respond. Now, a team of investigators have developed an integrated genomic approach that potentially could help physicians predict which patients with non-small cell lung cancer will respond to therapy with immune checkpoint inhibitors.

New iPSC Culture Medium Promises Weekends Off at Low Costs

Stem cell biologists rarely get weekends off. Seven days a week, their labs need someone to change the media keeping induced pluripotent stem cells alive. For many groups that use iPSCs, weekend lab visits — and expensive culture media — are considered inescapable facts of research.
In an effort to solve both problems, researchers at Northwestern University's Feinberg School of Medicine have developed a new human iPSC culture medium any lab can make from scratch at low cost and while taking weekends off.

EU Moves Toward Strict Controls on 3 Plastics Chemicals and PFBS

The European Union is taking aim at two chemicals used in polymer production, a phthalate, and a perfluorinated compound and its salts. All are now on a list of substances that are candidates for strict regulation.
If the chemicals on the list are selected for further controls, companies would only be allowed to use them if the European Chemicals Agency authorizes specific uses. ECHA says it added three of the substances to the list on Jan. 16 because they are reproductive toxics.
