While cryo-EM is revolutionizing structural biology, its impact on enzymology is yet to be fully demonstrated. The ketol-acid reductoisomerase catalyzes conversion of (2S)-acetolactate or (2S)-aceto-2-hydroxybutyrate to 2,3-dihydroxy-3-alkylbutyrate. We found that KARI from archaea Sulfolobus solfataricus is unusual in being a dodecamer, bispecific to NADH and NADPH, and losing activity above pH 7.8.

A Massachusetts General Hospital research team reports that several of the recently developed CRISPR base editors, which create targeted changes in a single DNA base, can induce widespread off-target effects in RNA, extending beyond the targeted DNA. Their report receiving advance online publication in Nature also describes engineering variant base editors that significantly reduced the incidence of RNA edits while also increasing the precision of on-target DNA editing.

The advance of science is something like the wandering of an explorer through an uncharted jungle. Often, the dense undergrowth can seem impenetrable, but at certain privileged moments, a clearing opens and an entirely new landscape emerges.
Something like this is occurring in the field of biology with the recent discovery of powerful techniques for intervening in the genetic code of life.

A concern that has plagued CRISPR-Cas9 technology from the beginning is the possibility, and unknown effect, of off-target effects—DNA mutations that result from CRISPR-Cas9 editing that are not in the targeted area. In order to understand the effect of off-target changes, they must first be detected. A new universally applicable approach for unbiased off-target identification has been developed.

The Scientist
Sequencing can identify mutations linked to autism even before a child's birth — especially in cases where doctors suspect problems, two new studies suggest.
In the studies, scientists sequenced fetal DNA only when ultrasounds revealed atypical development of limbs or other organs, and they gave families only the results that seemed to explain those problems.

Discovering new biological targets is a critical part of our ongoing battle against diseases. Over the years, scientists have made impressive progress towards the understanding of biological systems, constantly identifying novel targets. The structural diversity of these targets requires a broad range of different therapeutic agents.
"Small synthetic molecules are still key players, but biomolecules such as peptides, proteins and oligonucleotides have become an important area of research," says Professor Jérôme Waser.

Polymers have previously been made in the presence of living cells, for such purposes as encapsulating the cells or engineering their surfaces. But people haven't known whether synthetic polymers could be made inside cells, given the presence of free-radical scavengers. Mark Bradley and coworkers at the University of Edinburgh now show that radical polymerization can indeed happen inside living cells.

Cryogenic-Electron Microscopy has been a game changer in the field of medical research, but the substrate, used to freeze and view samples under a microscope, has not advanced much in decades. Now, thanks to a collaboration between Penn State researchers and the applied science company Protochips, Inc., this is no longer the case.
"The traditional type of grid hasn't changed much since the inception of cryo-EM, while materials science has changed vastly," said Deb Kelly, a professor of biomedical engineering at Penn State.
